The effects of high-fat diet on alcohol-induced liver injury were assessed using an intragastric intubation model. Urine alcohol content was monitored daily. Mice were evaluated for endoplasmic reticulum stress response as well as lipid and one-carbon metabolism. There were 2 cohorts: (1) mice fed high-fat diet only; (2) mice fed high-fat diet plus alcohol.
Procedures conducted:
• metabolic panel
Alanine transaminase (serum) after 4wk regimen of ethanol vs. control, both groups on high-fat diet. 24h fast.
• alcohol quantification
Alcohol concentration in blood and urine after 4 wk regimen of ethanol vs. control, both groups on high-fat diet. 24h fast.
• organ weights
Liver weight after 4wk regimen of ethanol vs. control, both groups on high-fat diet. 24h fast.
• histopathology
Liver histopathology, injury after 4wk regimen of ethanol vs. control, both groups on high-fat diet. 24h fast.
• metabolite quantification
Homocysteine concentration after 4wk regimen of ethanol vs. control, both groups on high-fat diet. 24h fast.
• lipid profile
Hepatic triglycerides after 4wk regimen of ethanol and high-fat diet. 24h fast.
• RNA expression profiling
Liver gene expression after 4wk regimen of ethanol and high-fat diet. 24h fast. mRNA abundance relative to the control (high-fat only) group.
