To identify genes and pathways that contribute to obesity-induced type II diabetes,
we constructed an F2 intercross between the diabetes-resistant C57BL/6J and diabetes-susceptible BTBR T+ tf/J mouse strains.
All F2s were genetically obese through introgression of the leptin mutation (Lep ob/ob) and all were sacrificed at 10 weeks of age.
We conducted genome-wide gene expression profiling in six tissues: adipose, gastrocnemius muscle, hypothalamus, pancreatic islets, kidney, and liver.
Gene expression was assayed with custom Agilent two-color ink-jet microarrays. Mice were genotyped with the Affymetrix 5K GeneChip.
